This is the final quest for the Dark Brotherhood questline.
You will have to do the reputation-building quests to move up the ranks. I believe the final quest unlocks at Dark Brotherhood Level 8.
You could easily knock this out in a day. The questline isn't long. However, you will have to keep picking up the quests from the contract book just to the right when you enter the Sanctuary.
